General Description:
Physiological analysis of growth involves the direct or indirect observation of variables attributable to normative functioning of systems and subsystems in living organisms.The study of the growth of bacterial cultures constitute an important method in Microbiology, specially for microbial physiology studies because bacteria’ composition and behavior are greatly influenced byenvironmental factors and growth conditions. Microbiologists have frequently stated that we should give more attention to the media and culturing conditions The purpose of this project is to determine the optimal growth conditions for gram-negative non-endospore bacteria Escherichia coli and Pseudomonas aeruginosa to further perform structural spectroscopic analysis E. coli has become the bacterium of choice for microbiology research and gene function analysis. In their optimum development medium, they also reproduce relatively quickly, doubling in number every 20 minutes. On the other hand, Pseudomonas sp. are often often labelled as “slow-growing pigmented bacteria”. In ideal conditions and in rich medium, P. aeruginosa takes 16–24 hours to grow. Growth on minimal medium can take longer depending on the nutrition sources available and the carbon source’s concentration. The bacterial growth curve and optimal growth conditions will be determined spectrophotometrically by optical density. The insights will be applied to future research including bacterial detection and discrimination using spectroscopic techniques.
